Hepatoprotective effect of clam (Corbicula fluminea) protein hydrolysate on alcohol-induced liver injury in mice and partial identification of a hepatoprotective peptide from the hydrolysate

In the current research, edible meat of freshwater clam Corbicula fluminea was subjected to enzymatic peptide extraction using animal protease. The protein hydrolysate (CPH) contained 45.78% essential amino acids and 44.97% hydrophobic showed high alcohol dehydrogenase (ADH) activation in vitro. Four fractions were divided from CPH by ultrafiltration further investigated for their hepatoprotective effects on alcohol-induced injury mice. 2.5-5 kDa fraction (CPH-3) best effect preventing hepatic damage caused alcohol, as revealed reduced serum aspartate transaminase (AST) alanine (ALT) activities malondialdehyde (MDA) level, marked decrease triglyceride (TG) contents, enhanced ADH, aldehyde (ALDH), glutathione (GSH) activities. Furthermore, histopathology CPH-3-treated mice less tissue compared alcohol- or other CPHs-treated Hence, CPH-3 purified consecutive chromatographic techniques with gel filtration reverse-phase high-performance liquid chromatography. One peptide, KKLAGFLRYYS (1330.68 Da), identified LC-MS/MS, which exhibited strong ADH activity. Results suggested that could be a potential nutraceutical facilitating metabolism ameliorating early liver induced acute exposure is one main bioactive components CPH.
